OR16-4 Genomic Interrogation of the Phytoestrogen Genistein in Ovarian Cancer Cells

چکیده

Abstract Although studies have shown beneficial effects of the phytoestrogen genistein on reducing ovarian cancer risk, little is known at molecular level regarding how inhibits growth, with estrogen receptor beta (ERb), a hormone-regulated transcription factor, receiving bulk attention. When endogenous and exogenous ligands engage ERb, binds to regulatory regions in genome as enhancers, where it associates coregulatory proteins, increases histone modifications associated active promotes chromatin looping, stimulates enhancer production RNAs, ultimately regulates target gene transcription. Since signaling pathways converge transcriptome level, an unbiased approach needed examine acts suppress growth beyond ERb. We previously used global run-on sequencing (GRO-seq), high-throughput method that identifies location orientation all transcriptionally RNA polymerases across genome, identify enhancers based enrichment aforementioned genomic features. To determine responses upon treatment cells, we PRO-seq characterize landscape OVCAR3 OVCAR4 cell lines treated genistein. The advantage this lack priori assumptions which are regulated by genistein, since allows pathway determination universe enhancers. Ultimately, will allow us uncover ERb ligand change cells.

Genistein, a soy phytoestrogen, prevents the growth of BG-1 ovarian cancer cells induced by 17β-estradiol or bisphenol A via the inhibition of cell cycle progression.

An endocrine disrupting chemical (EDC) is a global health concern. In this study, we examined the effects of genistein (GEN) on bisphenol A (BPA) or 17β-estradiol (E2)-induced cell growth and gene alterations of BG-1 ovarian cancer cells expressing estrogen receptors (ERs). In an in vitro cell viability assay, E2 or BPA significantly increased the...

Permanent Oviduct Posteriorization after Neonatal Exposure to the Phytoestrogen Genistein

BACKGROUND Preimplantation embryo loss during oviduct transit has been observed in adult mice after a 5-day neonatal exposure to the phytoestrogen genistein (Gen; 50 mg/kg/day). OBJECTIVE We investigated the mechanisms underlying the contribution of the oviduct to infertility. METHODS Female mice were treated on postnatal days 1-5 with corn oil or Gen (50 mg/kg/day). We compared morphology,...

The Phytoestrogen Genistein Affects Zebrafish Development through Two Different Pathways

BACKGROUND Endocrine disrupting chemicals are widely distributed in the environment and derive from many different human activities or can also be natural products synthesized by plants or microorganisms. The phytoestrogen, genistein (4', 5, 7-trihydroxy-isoflavone), is a naturally occurring compound found in soy products. Genistein has been the subject of numerous studies because of its known ...

The phytoestrogen genistein enhances osteogenesis and represses adipogenic differentiation of human primary bone marrow stromal cells.

In the present study, we investigated the role of the phytoestrogen genistein and 17beta-estradiol in human bone marrow stromal cells, undergoing induced osteogenic or adipogenic differentiation. Profiling of estrogen receptors (ERs)-alpha, -beta1, -beta2, -beta3, -beta4, -beta5, and aromatase mRNAs revealed lineage-dependent expression patterns. During osteogenic differentiation, the osteoblas...
